邻近闪烁分析

概要:The use of appropriate radioisotopes and scintillant incorporated microbeads allows the scintillation proximity assay (SPA) to become truly homogeneous and high-throughput. As there are ways of capturing radiolabelled molecules either directly or via a binding partner, the assay could be used for sensitive detection of enzymatic process as well as protein-protein interaction. For the former, we have developed a SPA assay for a methyltransferase. The ability of the enzyme in putting a radiolabelled methyl group onto a biotinylated peptide is tested in the presence of an inhibitor. SPA is relatively a versatile technique and has broad assay application, Biortus looks forward to utilise the full potential of this assay technology for our clients.

邻近闪烁分析(SPA)选用合适的放射性同位素和结合了闪烁剂的微珠,使其实现了真正的均一性和高通量。 由于有许多直接或者间接(固定配体)捕获放射性标记分子的方式,该方法可以灵敏的检测酶促反应过程以及蛋白质-蛋白质的相互作用。对于前者,我们已经开发了甲基转移酶的SPA分析方法, 在抑制剂存在的条件下测试酶将放射性标记的甲基转移到生物素化多肽上的能力。 SPA是一种相对多样化的技术,并且具有广泛的分析应用,佰翱得期待利用这种分析技术为我们的客户提供满意的服务。
